What researchers have looked at

• Discovery (2015): a mitochondrial-encoded peptide acting on skeletal muscle, activating AMPK by inhibiting the folate cycle; treated mice resisted diet-induced obesity and insulin resistance^[1].
• Ageing and physical capacity (2021): improved performance in young, middle-aged and old mice, and increased healthspan when intermittent treatment began late in life^[2].
• Muscle wasting (2024): in cast-immobilised mice, muscle loss fell from about 15% to about 5% with MOTS-c, alongside lower inflammatory cytokines and less intramuscular lipid^[3].
• Limits: in people only endogenous MOTS-c has been measured — plasma levels were similar in lean and obese adults but tracked insulin resistance in the lean group^[4] — and reviews record no developed clinical use^[5].

A 16-amino-acid peptide (MRWQEMGYIFYPRKLR) encoded inside the mitochondrial 12S rRNA gene and described in 2015. In mice it activated AMPK through the folate–purine pathway and was studied for insulin resistance, exercise capacity and muscle wasting (PMIDs 25738459, 33473109, 38170165).

Is the MOTS-c pen for human use?

No. MOTS-c has no approved product, and synthetic MOTS-c has never been given to people in a published trial; the human studies measured the body’s own peptide (PMID 29593067). The pen is for laboratory research only.
