What researchers have looked at

  • Metabolism in mice (2015): the discovery study found that MOTS-c inhibits the folate cycle and de-novo purine synthesis, activating AMPK, and that treated mice were protected from age-dependent and diet-induced insulin resistance and from diet-induced obesity[1].
  • Nuclear signalling (2018): under glucose restriction the peptide moved into the nucleus in an AMPK-dependent way and regulated antioxidant-response genes with NRF2[2].
  • Ageing mice and exercise (2021): MOTS-c enhanced physical performance in 2-, 12- and 22-month-old mice, and in humans exercise raised endogenous MOTS-c in muscle and blood[3].
  • Limits: the human evidence is observational — for instance plasma MOTS-c in ten lean and ten obese adults[4] — and reviews state that no clinical application has been developed[5].
